Wowser....
What a choice.

At this early hour, and with my sleep deprivation...

I would have to say Josey Wales...

There are a few that I watch every time they come on such as Pale Rider, Wyatt Earp, Tombstone but I think I have to go with Josey Wales as my favorite.

By far, my favorite. The ultimate American “morality play”.

I don't typically watch westerns, but I'd have to say High Noon is probably my favorite. It's the only western I own on DVD.

I also like:

Open Range
Tombstone
Wyatt Earp
(I actually preferred Wyatt Earp to Tombstone)
The Shootist
Unforgiven

The Searchers.
Not merely a great Western, but a great movie by any standards.

Yup. I agree. Leone + Morricone + the best acting talent.

Here's the final shootout between Chuck Bronson and Jane Fonda's pappy:

Yep, don't think it's possible to name just one as a favorite. I like many (most) of those mentioned so far.

I would add:

Butch Cassidy and the Sundance Kid and

True Grit (I like both, but give the nod to the Coen Brothers version)
