The three that come to mind for me have already been mentioned, but I'll second them:

Hendrix: (Dylan's) All Along The Watchtower
Clapton: (Bob Marley's) I Shot The Sheriff
CSNY: (Joni Mitchell's) Woodstock

EDIT: mikeyspizza below reminds me of Cocker; he did so many great covers it's hard to know where to start. In addition to mikey's choice, think of Cry Me A River, for example. I love the classic renditions, too (Julie London, for instance), but his take is amazing. Just look up the YouTube video from the Mad Dogs tour.

And, yeah, Johnny Winter doing Highway 61. Good stuff.

And, come to think of it, turnabout is fair play; Derek and the Dominoes doing Hendrix's Little Wing
