I agree, early in their careers, they were quite good in being able to play different characters. Travis Bickle, Vito Corleone, and Jake LaMotta- that is quite a diverse line up for De Niro, whose acting has become a parody of himself in the later part of his career. But is seems the majority of movie stars/actors throughout history fit the "one character" premise of this thread, they simply can't deviate from their physical looks and traits, voice, mannerisms, etc. As mentioned, John Wayne, Clint Eastwood, both made excellent films. But also Cary Grant, Humphrey Bogart, Jimmy Stewart, Spencer Tracy, guys who seem to mostly play themselves on screen. Will Farrell immediately came to mind. He's so repetitive that I can't find humor in his performances. Adam Sandler has actually has demonstrated some decent chops in serious roles. In addition to the ones already mentioned, I thought he was great in Spanglish and in Hustle playing a scout for the 76ers. "You know, Scotland has its own martial arts. Yeah, it's called Fuck You.
