The directions for my holsters which have retention screws indicate that they should tightened so the force needed to draw your pistol is to your liking. I have adjusted mine through a wide range of tension to see what I preferred. When the tension was high, it was pretty tough getting the pistol in and out. Short answer, I don't think it is likely you will tighten them to the point there will be any damage to the holster.
