My wife and I have been on the trail 3 or 4 times. She has a good time and doesn't even like bourbon.
I would recommend Jim Beam as they have the largest campus with the most to do. After that I like going through makers mark. All the distilleries have something unique to go for.
If you watch your time you'll probably only get about 3 of them done in a day.
If you have the time, try going through Independant Stave Company. They are a cooperage that makes barrels for a variety of makers in and out of the area.

lugerguardsI stayed in Louisville the 2 times I went down to the trail. BT is a must, Four Roses is also cool. I had fun at Bourbon Row in downtown Louisville. I also have done AngelsENvy. Wanted to do rabbit hole but never got there. The Louisville Omni is top class, great bars there along with food. I enjoy Doc Crows when I am in town also.

ohhh and if your in downtown Louisville, avoid Fourth street.

95flhrI did the Makers ambassador tour in November. As said above tours fill up quickly at about all the distilleries.
Buffalo Trace is my Favorite, and I also enjoy many of the smaller distilleries that you can find.
