Old Clint Eastwood/Sandra Locke flick called "Bronco Billy". Eastwood played Bronco Billy, running a travelling wild west show. Locke was a rich girl running away from an arranged marriage.

Eastwood says something about being in prison:

Locke: You were in prison?
Eastwood: Yeah.
Locke: For what?
Eastwood: For 6 years.
Locke: No, what did you do?
Eastwood: Murder.
Locke: Oh my God, what happened?
Eastwood: I came home one afternoon and found my wife in bed with my best friend.
Locke: What did you do?
Eastwood: I shot her.
Locke: Shot her? What about him?
Eastwood (obviously confused for a moment): Well, he's my best friend.

Some class movies listed here, but the winner can only be, Dirty Harry. Yeah, not so many lines, but it's without a doubt THE classic line of all time.
